Current research is focused on the structure determination of transcription factors (TFs) involved in the early stages of stem cell development. We are leveraging upon GIS genomic discovery groups to look at novel sites of binding as well as newly annotated proteins. Although we seek to determine high-resolution structures employing X-ray crystallography as the ultimate goal for several TFs, we also apply a series of other techniques including EMSA and pull-down assays to better characterize and analyze protein-protein as well as protein-DNA interactions. Using a combination of techniques we are increasingly moving towards working with larger complexes. Our ultimate aim is to understand stem cell regulation at the molecular and atomic levels.
